“Understanding MAPK Signaling in Dynamics“

Every living organism constantly senses intrinsic and environmental signals, and reprograms cellular responses to grow and maintain the lives.  Mitogen activated protein kinase (MAPK) cascades are evolutionarily conserved and extremely versatile signaling modules from plants to humans. In Arabidopsis, MPK4 and MPK3/6 modules govern a wide range of cellular programs in stress and immune responses as well as in growth and development. However, it remains challenging to elucidate definitive roles of MAPK during specific signaling pathways in multicellular organism, as the genetic mpk mutants often confer complexity to genotype-to-phenotype problem. We integrated systematic network and transcriptome analyses with time-resolved and quantitative chemical perturbations of genetically engineered MAPKs for signaling interrogation in Arabidopsis thaliana. We demonstrate specific and convergent signal processing through the core MAPK modules in diverse stimuli responses as well as threshold activity-based discrimination of MAPK regulation in multiple biological programs. Chemical genetic-based systems analyses in precise time and dosage open new possibilities to discover and reconstruct MAPK signaling networks in broad contexts of whole organisms.
